Mechanism of action

Novfloxcin acts primarily by inhibiting bacterial DNA gyrase and topoisomerase IV. These enzymes are essential for DNA replication, repair, and transcription in bacteria. By preventing these enzymes from functioning, novfloxcin disrupts the supercoiling of DNA, which is necessary for bacterial cell division and survival.

Pharmacodynamics

Novfloxcin exhibits concentration-dependent bactericidal activity against a wide range of gram-negative and some gram-positive bacteria. It demonstrates a time-dependent killing effect, which means that the efficacy of the drug increases with higher concentrations and prolonged exposure to the bacteria. Resistance to novfloxcin can develop through mutations in the target enzymes or through efflux pump mechanisms.

Pharmacokinetics

Novfloxcin is well absorbed after oral administration, with peak plasma concentrations typically reached within 1 to 2 hours. The drug has a bioavailability of approximately 70-80%. It is widely distributed throughout the body, including tissues and fluids, with a volume of distribution of about 1-2 L/kg. Novfloxcin is primarily excreted via the kidneys, with approximately 50-70% of the dose eliminated unchanged in the urine. The elimination half-life is approximately 6-8 hours, which may be prolonged in patients with renal impairment.
